SSH-Plates: The SSH-plates
contain cDNAs from Halina Knap’s laboratory, Clemson University. The clones
(776) were from subtractive DNA libraries of PI 437654 roots.
Among these clones, 444 clones (No. 1-12F to No. 9-3H) are from a forward
subtractive cDNA library, in which the mRNA from roots 10 hrs post infection
by SCN was used as tester and the non- infected root mRNA was used as driver.
The other 332 cDNA clones (No. Rq to No. R99) are from a reverse-subtractive
cDNA library, in which the non-infected root mRNA was used as tester and the
mRNA from roots infected 10 hrs with SCN was used as driver.
All these cDNAs were cloned in pGEM-T vector (Promega) and sequenced with
the T7 primer. |
